決明子萘并吡喃酮類(lèi)對(duì)照提取物研究及其在決明子藥材質(zhì)量控制中的應(yīng)用

[摘要] 建立以決明子萘并吡喃酮類(lèi)對(duì)照提取物為對(duì)照的決明子藥材質(zhì)量控制方法,探討對(duì)照提取物在中藥質(zhì)量控制中應(yīng)用的可行性。采用色譜分離技術(shù)制備決明子萘并吡喃酮類(lèi)對(duì)照提取物,以決明子苷B2、紅鐮霉素6OβD龍膽二糖苷及決明子苷C對(duì)照品為對(duì)照,對(duì)決明子萘并吡喃酮類(lèi)對(duì)照提取物進(jìn)行標(biāo)定。以已知含量的對(duì)照提取物為對(duì)照,建立決明子藥材HPLC含量測(cè)定方法,并與以對(duì)照品為對(duì)照的含量測(cè)定結(jié)果進(jìn)行比較,結(jié)果表明,2種含量測(cè)定方法無(wú)顯著差異。該結(jié)果為決明子萘并吡喃酮類(lèi)對(duì)照提取物在決明子藥材質(zhì)量控制中的應(yīng)用提供科學(xué)依據(jù),為中藥對(duì)照提取物替代單一對(duì)照品進(jìn)行中藥質(zhì)量控制奠定基礎(chǔ),為中藥質(zhì)量控制提供新的研究思路和模式。

Study on naphthopyrone reference extract and application on
assay of Semen Cassiae

[Abstract] A quality control method of Semen Cassiae was established by using naphthopyrone reference extract(NRE). Meanwhile, the feasibility about NRE replacing single component reference in quality control of traditional Chinese medicine was explored. After NRE of Semen Cassiae being prepared by chromatographic separation technology, we determined the three main components, cassiaside B2, rubrofusarin6OβDgentiobioside and cassiaside C. In the meantime, an HPLC analytical method, based on the NRE as standard substance, was developed to determinate the contents of three main components in Semen Cassiae. Ttest was used for comparison of the determination results of the two methods(single chemical component and NRE as reference substances, respectively), and the Ttest result demonstrated that there was no significant difference between the two methods. The results developed scientific basis for the application of NRE of Semen Cassia in the quality control, which could be applied for the quality control of traditional Chinese medicine using reference extract substituting single chemical reference, and provide a new research model for the quality control of Chinese medicine.

中藥質(zhì)量控制與評(píng)價(jià)是中藥研究領(lǐng)域的重要科學(xué)問(wèn)題,現(xiàn)行中藥質(zhì)量控制方法主要以單一化學(xué)成分為對(duì)照品,從定性和定量角度對(duì)中藥及復(fù)方進(jìn)行質(zhì)量控制。這種以單一化學(xué)成分表征中藥整體質(zhì)量的模式無(wú)法真正控制和評(píng)價(jià)中藥質(zhì)量,與中醫(yī)藥整體觀亦不相符,在一定程度上阻礙了中藥現(xiàn)代化、國(guó)際化的進(jìn)程。中藥成分的復(fù)雜性決定了以單一成分或指標(biāo)成分為對(duì)照難以客觀評(píng)價(jià)中藥質(zhì)量,然而,多指標(biāo)成分的質(zhì)量評(píng)價(jià)模式卻受到中藥化學(xué)對(duì)照品分離難度大、單體穩(wěn)定性差或?qū)嶒?yàn)成本高等因素的制約[1]。針對(duì)這一問(wèn)題,有研究者[23]提出了一測(cè)多評(píng)法,該方法可在一定程度上解決中藥多成分同時(shí)定量的一些問(wèn)題,但同時(shí)該方法在相對(duì)校正因子的定量、指標(biāo)成分的色譜峰定性,尤其是方法的適用性和耐用性方面尚存在一些問(wèn)題。因此,建立符合中藥多成分特點(diǎn)的新方法是解決中藥質(zhì)量控制問(wèn)題的有效途徑。
中藥對(duì)照提取物是指主要化學(xué)成分及其含量明確、穩(wěn)定性好、純度較高的中藥提取物,其化學(xué)成分組分比例相對(duì)固定,指標(biāo)性成分含量已采用相應(yīng)的對(duì)照品進(jìn)行標(biāo)定,可以實(shí)現(xiàn)同時(shí)對(duì)多個(gè)組分進(jìn)行定性和定量分析,不但制備方法相對(duì)簡(jiǎn)單、成本較低,而且后續(xù)質(zhì)量控制過(guò)程相對(duì)簡(jiǎn)潔、實(shí)驗(yàn)步驟相對(duì)簡(jiǎn)便,用于中藥材多成分分析,可節(jié)約資源,降低檢驗(yàn)成本[4]。如2015年版《中國(guó)藥典》收載了銀杏葉片劑2 種可供選擇使用的含量測(cè)定方法,若使用化學(xué)對(duì)照品進(jìn)行含量測(cè)定,則需使用槲皮素、山柰素、異鼠李素等7種化學(xué)對(duì)照品,若使用已標(biāo)定的2種銀杏葉對(duì)照提取物進(jìn)行含量測(cè)定,成本將大大降低,同時(shí)可以減少單體對(duì)照品的使用,減少實(shí)驗(yàn)步驟和實(shí)驗(yàn)成本[5]。因此,中藥對(duì)照提取物的研發(fā)與應(yīng)用不僅可以解決中藥質(zhì)量控制中成本高、穩(wěn)定性差、重現(xiàn)性差等諸多問(wèn)題[68],而且為更科學(xué)、合理、系統(tǒng)的開(kāi)展中藥質(zhì)量控制,提供新的研究方法和研究模式。
