沒(méi)食子提取物體內(nèi)外抗炎作用研究

[摘要] 目的 研究沒(méi)食子提取物體內(nèi)外抗炎作用。 方法 體內(nèi)以二甲苯致小鼠耳腫脹、角叉菜膠致大鼠足趾腫脹實(shí)驗(yàn)觀(guān)察其抗炎作用;體外觀(guān)察沒(méi)食子提取物對(duì)RAW264.7細(xì)胞生長(zhǎng)曲線(xiàn)和活力以及吞噬中性紅能力的影響;采用酶聯(lián)免疫吸附測(cè)定法(ELISA)檢測(cè)沒(méi)食子提取物對(duì)脂多糖(LPS)刺激的RAW264.7細(xì)胞分泌腫瘤壞死因子α(TNF-α)的影響。 結(jié)果 與模型組比較,沒(méi)食子提取物能顯著減輕小鼠的耳腫脹度和足趾腫脹(P < 0.05或P < 0.01);20、40 μg/mL沒(méi)食子提取物能活化RAW264.7巨噬細(xì)胞,促進(jìn)RAW264.7細(xì)胞增殖,并能增強(qiáng)其吞噬功能;25、50 μg/mL沒(méi)食子提取物對(duì)LPS誘導(dǎo)的RAW264.7細(xì)胞分泌TNF-α有明顯的抑制作用(P < 0.01)。 結(jié)論 沒(méi)食子提取物具有一定的抗炎作用,可能與抑制促炎介質(zhì)TNF-α的分泌有關(guān)。

[Abstract] Objective To study the anti-inflammatory effects of Turkish galls extract in vivo and in vitro. Methods The anti-inflammatory effects in vivo were observed through the experiments of mice ear swelling induced by xylene and rats toe swelling induced by carrageenan; the effects of Turkish galls extract for the growth curve and vitality of RAW264.7 cells and phagocytosis of neutral red were observed in vitro; the effects of Turkish galls extract for the secretion of tumor necrosis factor α (TNF-α) in RAW264.7 cells stimulated by lipopolysaccharide (LPS) were detected by enzyme linked immunosorbent assay (ELISA). Results Compared with model group, Turkish galls extract could significantly reduce the degree of ear swelling and toe swelling (P < 0.05 or P < 0.01). 20, 40 μg/mL of Turkish galls extract could activate RAW264.7 macrophages, the proliferation of RAW264.7 cells was promoted and their phagocytosis was enhanced. 25, 50 μg/mL of Turkish galls extract had significant inhibitory effects for the secretion of TNF-α in RAW264.7 cells stimulated by LPS (P < 0.01). Conclusion Turkish galls extract has certain anti-inflammatory effects, which may be related to the inhibition of secretion of pro-inflammatory mediator TNF-α.

沒(méi)食子是沒(méi)食子蜂科昆蟲(chóng)沒(méi)食子蜂Cynips gallae-tinctoriae Oliv.的幼蟲(chóng),寄生于殼斗科植物沒(méi)食子樹(shù)Quercus infectoria Oliv.幼枝上所產(chǎn)成的蟲(chóng)癭。沒(méi)食子性寒,味苦澀,具有固澀、收斂、燥濕、止血、消炎的作用[1-2],是維吾爾醫(yī)常用藥材,具有健齒固齦、清血止血、消炎和防腐之功效,被用于治療潰瘍性結(jié)腸炎、口腔潰瘍等疾病,療效顯著,臨床上其具有明顯的抗炎、鎮(zhèn)痛作用[3-4]。藥理學(xué)研究表明,沒(méi)食子具有提高機(jī)體非特異性免疫功能,清除自由基和抑制脂質(zhì)過(guò)氧化損傷等作用[3-4]。但其系統(tǒng)的抗炎作用報(bào)道較少,本實(shí)驗(yàn)通過(guò)研究沒(méi)食子提取物的體內(nèi)外抗炎實(shí)驗(yàn),體內(nèi)試驗(yàn)觀(guān)察沒(méi)食子提取物對(duì)二甲苯致小鼠耳腫脹、角叉菜膠致大鼠足趾腫脹的影響,體外試驗(yàn)觀(guān)察沒(méi)食子提取物對(duì)RAW264.7細(xì)胞生長(zhǎng)曲線(xiàn)和活力以及吞噬中性紅能力的影響;進(jìn)一步觀(guān)察采用酶聯(lián)免疫吸附測(cè)定法(ELISA)檢測(cè)沒(méi)食子提取物對(duì)脂多糖(LPS)刺激的RAW264.7細(xì)胞分泌腫瘤壞死因子α(TNF-α)的影響,探討其抗炎作用機(jī)制,為臨床用藥提供依據(jù)。
1 材料與方法
1.1 藥品
沒(méi)食子水提物(批號(hào):20161112),藥材購(gòu)自新疆奇康哈博維藥有限責(zé)任公司。由新疆維吾爾醫(yī)藥研究所希爾艾力主任藥師鑒定為正品。提取方法:取沒(méi)食子,粉碎成粗粉,加8倍量水浸泡1 h后,煎煮3次,每次0.5 h,合并煎液,濾過(guò),濾液真空干燥得浸膏,稱(chēng)重,得率為75%。
